Sherlock and Mycroft as the angels Kabniel and Ashriel respectively, two of the angels invoked to cure stupidity. (Ashriel also has the task of separating souls from bodies in the moment of death).
Sherlock 'knowing Ash' is a reference to him knowing Ashriel (however if you go with the Supernatural crossover version, is him knowing Ash, the computer guy who managed to hack Heaven).
When Sherlock says he might be on the side of the angels but he's not one of them, he's lying through his pretty teeth. (Or, he's using the distinction of 'I'm a fallen angel, I don't count')
